OK, when I add a new frequency in Transedit, it makes changes to the relevant ini file in transponders subfolder of the configuration folder, but it does not change the ini file in the transponders subfolder of Program Files/DVBViewer- what is the purpose of this other transponders folder and is it necessary for me to copy the changed ini file from the configuration folder to this other location when making changes with transedit?

It's just a backup of the originally installed transponder lists. They can't be changed there anyway except with admin rights. In a nutshell: One folder is the installation, the other one is for you and you can do there what you want.

No.

The only relevant location is the configuration folder.

..imho it's one of the most common errors that users put changed transponder.inis into the original installation folder and then wonder why a scan doesn't yield the wanted (HD) channels :(

I have copied the ini from the config folder to program files transponder folder, why will this cause errors if it is only a backup? FWIW I am trying to scan 10935V (Astra 2D/28.2) which has just changed to S2.
